  • Great exchanges during #CRS2024 with Adèle Rakotonirina, PhD including the transition from University to Industry. Surely a lot to learn from both perspectives to improve communication and knowledge and make such transitions easier.

    Adèle Rakotonirina obtained her PhD in Pharmaceutical Sciences in June 2023 at the University of Geneva. She worked on a novel oral formulation for fecal microbiota transplantation. During her PhD, she has discovered a passion for science communication by participating to "Ma thèse en 180 secondes" and wanted to work in a role that has this aspect. She has now transitioned to the industry as a Junior Business Development Manager. She uses her scientific knowledge and communication skills every day to connect with all kinds of scientists with innovative projects. She participated in the takeover at CRS 2024 to talk about her transition to the industry and presented the different career possibilities after university.

    Kicked off the first day of Controlled Release Society (CRS) #CRS2024 in Bologna! The workshop about Oral Drug Delivery organized by MeltPrep was very insightful with talks reminding us of the basics of oral formulations and promoting collaborations from actors in academia and industries to give the best solutions to patients. It was exciting to learn about innovative technologies, modeling programs and the effect of food intake on drug bioavailability, among others! Have look at "A linear scale-up approach to fluid bed granulation" by Victor Emenike, Dr. Ivonne Kulla, Martin Maus, Andrea Staab and Daniela Schröder and learn more about the "Maus factor" too. Fluid bed granulation (FBG) is used extensively in the pharmaceutical industry and it is known to be a complex process, because the final product quality of the FBG process is determined by a complex interplay between the process parameters, fluid dynamics, and material properties. Due to this complexity, the FBG process is inherently nonlinear and as such difficult to scale-up. The field of chemical engineering has shown that complex nonlinear processes can be assumed to be linear under limiting conditions. We leverage this idea and present a linear scale-up approach (LiSA) to the FBG process. We derive the key LiSA equation from first principles, and then use it in combination with the similarity principle for scale-up purposes. Furthermore, we present a novel regression-based LiSA. The regression-based LiSA is founded on the hypothesis that there is a linear relationship between the moisture content and a scaling parameter called the Maus factor. This hypothesis is based on our experience and it is shown to be plausible due to high 𝑅2 values ranging from 0.86 to 0.98. Moreover, we successfully demonstrate that LiSA is effective under typical industrial process settings by applying it to two different formulations during pharmaceutical drug product development. We at Glatt Pharmaceutical Services - CDMO Unlimited just love it - why? Because it is an versatile, efficient, scalable and high quality process when it comes to pellets and especially when it comes to apply functional coatings. Learn more in the video by Glatt Group. Here are some key factors about the Wurster Process you might not know yet: Quality  One of the most significant benefits of the Wurster process is its ability to achieve a uniform and consistent drug layering but also coating application. The unique design of the system, where pellets are suspended in an upward airflow, ensures that the drug and coating solution is evenly distributed across each particle's surface. This uniformity is critical for developing uniform drug loading and applying functional coatings for a tailored and consistent drug release profile. This precision allows for improved therapeutic outcomes and better patient compliance, as medications can be tailored to release at specific times or in particular areas of the body. Scalability  Scalability is another strength of the Wurster process. It is adaptable to various production scales, from small laboratory batches to large commercial manufacturing runs, without compromising the quality of the drug layering and coating processes. This flexibility is invaluable for pharmaceutical companies as they transition from development to full-scale production. Versatility  The versatility of the Wurster process extends to the range of coating materials it can handle. Polymers, sugars, waxes, and other substances can be used, allowing for customization of the coating's properties such as solubility, permeability, and mechanical strength. This adaptability helps in meeting diverse formulation requirements. The process can run with suspensions and solutions based on aqueous or organic solvents. Efficiency  Efficiency is also a hallmark of the Wurster process. The controlled environment within the fluid bed system minimizes material loss, ensuring that most of the coating solution is applied to the pellets rather than being wasted.
