✍️Discovering Mediator's role in Cell-Division - possibility of 'controlling' cell division : '.. research team at Umeå University has discovered how the Mediator, a protein complex in the cell nucleus, can bind to DNA and interact with another protein complex, Lsm1-7, to regulate the production of proteins that make up the ribosomes. The study shows that when cells grow too densely, cell division slows down. When this happens, the mediator moves to the end of the genes where it interacts with Lsm1-7. This has the dual effect of both slowing down the reading of the genes and interfering with the maturation of mRNA. This, in turn, leads to a reduced production of ribosomal proteins and thus a slower cell division. I am happy to announce my PhD research describing the role of LARP4 in regulating nuclear-encoded mitochondrial mRNAs is finally published and available online. Title:  LARP4 Is an RNA-Binding Protein That Binds Nuclear-Encoded Mitochondrial mRNAs To Promote Mitochondrial Function Abstract:  Mitochondria-associated RNA-binding proteins (RBPs) have emerged as key contributors to mitochondrial biogenesis and homeostasis. With few examples known, we set out to identify RBPs that regulate nuclear-encoded mitochondrial mRNAs (NEMmRNAs). Our systematic analysis of RNA targets of 150 RBPs identified RBPs with a preference for binding NEMmRNAs, including LARP4, a La RBP family member. We show that LARP4’s targets are particularly enriched in mRNAs that encode respiratory chain complex proteins (RCCPs) and mitochondrial ribosome proteins (MRPs) across multiple human cell lines. Through quantitative proteomics, we demonstrate that depletion of LARP4 leads to a significant reduction in RCCP and MRP protein levels. Furthermore, we show that LARP4 depletion reduces mitochondrial function, and that LARP4 re-expression rescues this phenotype. Our findings shed light on a novel function for LARP4 as an RBP that binds to and positively regulates NEMmRNAs to promote mitochondrial respiratory function. Our latest paper presents a systematic, large-scale computational study assessing cancer cell transit through complex a 3D geometry. We show that small variations in cellular properties akin to natural heterogeneity found across cell populations or cell lines can lead to significant differences in transit and flow force exposure. Junyu Nan and Sayan Roychowdhury's research article entitled "Investigating the Influence of Heterogeneity Within Cell Types on Microvessel Network Transport" was published in Cellular and Molecular Bioengineering this week. This work highlights the importance of considering cell heterogeneity in biophysical studies. It suggests that slight variations in cellular characteristics (e.g., radii or stiffness) can impact the fluid conditions the cell is exposed to and subsequently alter transport. The researchers approach modified #nanopore sequencing, a cutting-edge technique for deciphering #genetic material, to make the procedure simpler and maybe approachable. How does a mutation in a protein contribute to an observed or desired phenotype? To better understand this central question of biophysics, three Teams ScaDS.AI Dresden/Leipzig co-developed a new tool to analyze and visualize protein mutations in 3D: MutationExplorer. MutationExplorer combines easy accessibility and a large array of different tools to help researchers understand mutations of protein mutations and their effects on molecular structures. This helps researchers to better understand how mutations impact drug resistance, genetic diseases, and cancer. MutationExplorer features: An easy-to-use, browser-accessible interface for mutating protein models from the Protein Data Base Integrated tools for mutation effect analysis AI predictions of how mutations affect protein stability This tool was developed through a collaboration between the Leipzig University, Københavns Universitet - University of Copenhagen, Vanderbilt University, and the Hospital of the Bielefeld University, supported by the Federal Ministry of Education and Research, Germany and the Sächsische Staatsministerium für Wissenschaft, Kultur und Tourismus.
