A novel approach developed by scientist at the Broad Institute of MIT and Harvard allows researchers to attach precise location tags to individual cell nuclei for isolation in single-cell experiments. Known as Slide-tags, the new method builds upon a previously developed method known as Slide-seq. Slide-seq maps spatial patterns of genetic activity within tissues, but doesn’t reach single-cell resolution like the new Slide-tags method. A comprehensive map, or single-cell spatial distribution atlas, has been constructed for the entire macaque cortex. Scientists performed large-scale single-nucleus #RNA sequencing and spatial transcriptomic analysis of 143 macaque cortical regions. In doing so, they revealed the global distribution of 264 cell types and their marker genes. The work provides a guide for systematic analysis of cell-type distribution specificity and regional specificity within the cortex.

Researchers reveal that the activity of Snhg11, a gene found in the 'dark matter' of the genome, is critical for the function and formation of neurons in the hippocampus, specifically in an area critical for learning and memory. Researchers have discovered the gene is less active in brains with Down syndrome, potentially contributing to the condition's intellectual disabilities.  The study is the first evidence that a non-coding RNA plays a critical role in the pathogenesis of Down syndrome. #downsyndrome #intellectualdisability #neurons #learning #memory

New Publication Alert: Chung et al., Nature. 2024 The study was led by Changuk Chung, Ph.D., and Xiaoxu Yang, Ph.D., both from the laboratory of Joseph Gleeson, M.D., at the School of Medicine Department of Neurosciences and the Rady Children's Institute for Genomic Medicine. In the study, Chung et al., leverage cell-type resolved mosaicism to reveal clonal dynamics of the human forebrain. In addition to other cutting edge approaches, the research team utilized ResolveOME™-enabled DNA and RNA analysis from the same cell which allowed for the simultaneous identification of the cell type and genotype by the authors. New mouse brain atlas shows extraordinary diversity 🧠 The best, most comprehensive, and highly detailed map of a mammalian brain has dropped in an impressive set of nine papers from the BRAIN Initiative Cell Census Network (BICCN), a massive scientific collaboration funded by The National Institutes of Health. Profiling the spatial arrangements, functions, connectivity, gene expression, and neurotransmitter and neuropeptide activities in about 7 million separate brain cells, the new map is a benchmark reference atlas of the whole adult mouse brain.
