$\begingroup$ Combining your second suggestion with the "vines" example in the question to fit the "appear randomly/spontaneously" part: The "door" is a plant or fungus that grows over the door, and the "key" is a form of weedkiller - perhaps a magic sort, such as an enchanted nail that you drive into the main trunk, which would allow puzzles where you can see the door, but need to locate the "lock". $\endgroup$
