Proud to have co-organized #EuroSTEMPeers2024 with EuroSTEMPeers in Heidelberg! 🧬🔬💡 What an incredible two days of knowledge sharing and connection building. As BioLabs Heidelberg we aimed to create an environment where networking was just as valuable as the formal sessions - and the feedback has been phenomenal! Participants consistently highlighted how the program led to valueable insights around carreer, growth, entrepreneurship, personal development and networking Some highlights: • Engaging talks on academia-to-industry transitions • Interactive workshops on negotiation skills, stress management and design thinking • Thought-provoking panel discussions on entrepreneurship support and mid-career advancement • Invaluable networking sessions, including BioLab Tours (led by the BioLabs Heidelberg team (Ann-Kristin Mueller, Ornella Kossi, Heiner Sähr) It was inspiring to see so many connections being made between young professionals, researchers, industry professionals, and entrepreneurs. The energy and ideas exchanged during these interactions were truly the heart of the event. 𝐖𝐡𝐚𝐭 𝐢𝐟 the resolution, cost and accessibility of spatial transcriptomics could be enhanced? #OpenST, a high resolution, 3D spatial transcriptomics (ST) tool developed by Marie Schott, 𝐃𝐚𝐧𝐢𝐞𝐥 𝐋, Elena Splendiani and colleagues and published in Cell Press could be the answer to this challenging question. 𝐖𝐡𝐚𝐭 𝐝𝐢𝐝 𝐭𝐡𝐞𝐲 𝐝𝐨? ✅ Repurposed Illumina’s sequencing flow cell into ST RNA capture areas. They utilised standard lab equipment and optimised the technique to keep costs low per capture area. ✅ Tested the efficacy of Open ST on mouse brain tissue, primary human head and neck tumours and patient matched healthy/ metastatic lymph nodes. ✅ Developed an open access bioinformatics pipeline to generate virtual 3D tissue blocks by combining ST and morphological information (H&E staining) of successive tissue sections. 𝐖𝐡𝐚𝐭 𝐝𝐢𝐝 𝐭𝐡𝐞𝐲 𝐝𝐢𝐬𝐜𝐨𝐯𝐞𝐫? ✅ Compared to competitors, Open ST demonstrated high transcript capture and efficient library amplification, thereby making it cost effective as deeper sequencing could be achieved with fewer reads. ✅ Open ST could capture transcripts at sub cellular resolution in murine tissue and identify differential cellular heterogeneity in human tissues. When #benchmarked against an imaging based, targeted ST platform (Xenium, 10X Genomics), Open ST yielded comparable data. ✅ The reconstructed virtual 3D tissue blocks could be used to correlate gene signatures to biological function and cellular location, demonstrating the potential to identify biomarkers in 3D. While Open ST is exciting due to its resolution and cost effectiveness, it has several limitations that need to be overcome before this technology can become the standard in spatial transcriptomics. Anil is originally from #Cuttack, India. He has done master’s and PhD in #Physics from Pondicherry University, India, in the #Plasmonics research group lead by 𝐏𝐫𝐨𝐟. 𝐃. 𝐁𝐡𝐚𝐫𝐚𝐭𝐡𝐢 𝐌𝐨𝐡𝐚𝐧. Upon completing his doctoral studies, he worked as a postdoctoral fellow at the University of Birmingham. Prior to joining the BioMed X Institute, Anil was a #Humboldt Fellow in 𝐏𝐫𝐨𝐟. 𝐒𝐭𝐞𝐟𝐚𝐧 𝐌𝐚𝐢𝐞𝐫'𝐬 group at Ludwig-Maximilians-Universität München (Ludwig Maximilian University of Munich) in Munich, Germany. ❓ What if we could wirelessly monitor real-time physiological changes in the brain in a safe, minimally invasive way? Hanchuan Tang, Yueying Y., Zhen Liu, and colleagues address this in their paper published in Nature (Nature Portfolio) on the 5th of July 2024. 𝐖𝐡𝐚𝐭 𝐝𝐢𝐝 𝐭𝐡𝐞𝐲 𝐝𝐨? ✅ Designed an ultrasonic biosensor using a biodegradable, biocompatible metastructured hydrogel (metagel). ✅ Tested the functionality of this sensor compared to existing, standard clinical sensors in rat and minipig models. 𝐖𝐡𝐚𝐭 𝐝𝐢𝐝 𝐭𝐡𝐞𝐲 𝐝𝐢𝐬𝐜𝐨𝐯𝐞𝐫? ✅ In rats, the implanted metagel outperformed standard clinical intracranial probes at detecting changes in pressure and temperature. ✅ The position of the metagel in the rat brain remained stable for 4 weeks after implantation, with degradation occurring after 5 weeks. There was negligible immunoreactivity, indicating robust biocompatibility. The rats also exhibited no adverse health effects. ✅ In the pig model, under conditions closely resembling those in a clinic, the implanted metagel accurately measured changes in intracranial pressure and performed better than other standard sensors in terms of temporal resolution and sensitivity. This paper is an exciting development in the #biosensor field. The technology presented here has significant advantages over competing technologies as it is small, can integrate multiple signals, and is biodegradable.
