While your quads may never get a "complete" rest while using clipless pedals, they don't get any rest while using flats. One advantage of clipless pedals is that they allow a cyclist to shift the percentage of workload from quads to hamstrings, thus leading a cyclist to be fresher over the course of a longer ride.
